Environmental Monitoring In Microbiology Laboratory

·         PROCEDURE   :

           
Ø      The environmental monitoring of the area shall be carried out by settle plates.
Ø      The Personnel monitoring shall be carried out by taking finger dabs and garment samples using contact plates.
Ø      The Physical parameters like Temperature, Humidity and Differential pressure in the area shall be monitored.
·         MONITORING BY SETTLE-PLATE METHOD   
Ø      Enter the different areas following the appropriate gowning practices.
Ø      Expose the pre-incubated sterile media plates at different locations shown in annexure-I.
Ø      Collect the plates after completing exposure hrs and incubate at 20-25°C for 72 hrs followed by 30-35°C for 48 hrs.
Ø      The time of exposure, limits, frequency of monitoring applicable as per various grades and is attached in annexure II.
Ø      Record the counts in the format as mentioned in annexure III.
Ø      If the bio-burden is found out of the specified limit, proceed
·         PROCEDURE FOR PERSONNEL MONITORING
Ø      Transfer pre-incubated contact plates containing sterile SCDA media and the hand impressions and garment contacts of technician and helpers are taken at the place of operation.
Ø      After taking the samples close the lid of the petri –plate and write the name of the person, nature of work and date & time at the base of the plate.
Ø      Transfer those plates to the incubator having temperature set at 20-25°C for 72 hrs followed by 30-35°C for 48 hrs.
Ø      Observe the plates after completion of incubation period and note the observations in the format as mentioned in annexure IV.
Ø      If the bio-burden is found out of the specified limit, proceed.
Monitor, physical parameters like temperature, humidity and pressure differential at critical places, once a day, in microbiology laboratory in respective formats / Register.
